I tried runes a little, but I didn't use them besides speed and mana a few times, mainly because of only having a limited number per level. I was planning to use them more while unique hunting, but I didn't get to it. They are quite interesting I think, it's fun to set up an area like that. One thing is that I think you should be able to lay down more glyphs after you desanctify the ones existing, so you can use them for multiple vaults/uniques on a floor.
