When a ranger casts Resist Poison, they get the RPois flag at the bottom of the screen. Does that mean it stacks with poison resistance gained from equipping an item? Similar to how a potion of Resist Cold stacks with cold resistance gained from equipping an item?

Yep. Without rpoison from an item, it reduces poison damage to 1/3; if you have an item which already gives resistance to 1/3 damage taken, it increases that to 1/9. It works exactly the same as the 4 elements. -

Multiple perm and multiple temp do not stack
